Production of ¥ã-Glutamylcysteine by Immobilized Mixed Microbial System of Recombinant E. coli and Yeast

Abstract

|
y-Glutamylcysteine production by the immobilized microbial system of recombinant Escherichia colt and yeast was investigated. y-Glutamylcysteine was synthesized from L-glutamic acid and L-cysteine in the presence of ATP by the reaction catalyzed by y-glutamylcysteine synthetase. An immobilized mi¡©crobial cell system was developed for the efficient y-glutamylcysteine production. Recombinant Esche¡©richia coli and yeast were immobilized by alginate. Production of y-glutamylcysteine was better with the recombinant Escherichia colt for both the synthesis of y-glutamylcysteine and the ATP regeneration than the mixed system of recombinant Escherichia colt and yeast. The proper ratio of recombinant Esch¡©erichia colt to yeast was experimentary observed to be 1:4 in the mixed system. Although the immobi¡©lized system had the slower reaction rate, its reaction stability was increased by 10%.
